Identification of physical and chemical factors regulating gene expression and infectivity of the Lyme disease spirochetes

Borrelia afzelii is one of the main Lyme disease causative agents. It is known that differential gene expression during the Borrelia life cycle can relate to infectivity for mammals, including humans. However, just a little is known about what affects these changes in expression. Here, we investigate the effect of temperature and common blood nutrients on Borrelia afzelii differential gene expression and transmission efficiency while stimulating the ticks with chosen substances.
The Interaction of Borrelia Outer Surface Proteins with Tick Salivary Proteins

The aim of this thesis was to investigate the interaction of the salivary gland protein, Salp15, in relation to the outer surface protein C, OspC, using in silico analysis, as well as trying to amplify Salp15 from different tick species found in same geographical areas and observing the growth of Borrelia in the midgut and salivary glands of these different tick species in vitro.

We studied if the blockade of PD-1/PD-L1 axis by neutralizing antibodies influences the proliferation of T lymphocytes and cytokine milieu in imunological synapsis between murine dendritic cells and T cells in vitro.
Differentiating the adhesion properties of surface proteins between various Borrelia species

The aim of the thesis was to gain a better understanding of decorin binding proteins A and B (DbpAB) of various Borrelia species my different means. Overexpression of DbpA or DbpB in B. afzelii was addressed, examining phenotypic changes in mutants. Also, it was aimed to create shuttle vectors to complement DbpAB of different European Borrelia species as well as suicide vectors.
Growth kinetics of the Lyme disease spirochetes in vector ticks \kur{Ixodes ricinus} and \kur{Ixodes scapularis}

Growth kinetics of Borrelia afzelii CB43 in Ixodes ricinus and Ixodes scapularis were obtained. Based on this data, I. scapularis was marked as capable of acquiring B. afzelii infection. Growth kinetics of B. burgdorferi N40 in I. ricinus was obtained as well with the same outcome, I. ricinus was able to acquire B. burgdorferi infection. Following transmission experiments showed that I. ricinus as well as I. scapularis are able to acquire the infection by B. burgdorferi and B. afzelii. Moreover, we proved that both ticks are able to transmit the infection back to na?ve mice.
Understanding the interactions between Borrelia duttonii and the tick Ornithodoros moubata as well as the mammalian host at the in vitro and in vivo level

The aim of this Bachelor thesis was to study the interactions between the spirochete B. duttonii, which is a causative agent for relapsing fever in Africa, and its vector, the soft tick O. moubata. Several experiments were conducted including passaging, membrane feeding and keeping infected ticks at different temperatures. Microscopy experiments were carried out to visualize the presence of B. duttonii in mammalian blood cells and in the salivary glands of the O. moubata tick.
Adapting Genetic Methods For Transposon Mutagenesis And Fluorescent Markers In Borrelia

The aim of this Bachelor thesis was the examination and generation of different mutants in various Borrelia species. While several assays were used to examine aspects of B. burgdorferi transposon mutants, genetic tools were applied in order to generate fluorescent markers in B. afzelii and a vector construct in B. burgdorferi. This work should provide an overview of various genetic methods and their methodology that have been developed in Borrelia bacteria.

The thesis was written with the intention to bring together cutting-edge imaging methods and applications in order to illustrate how imaging can answer pathogenesis-related questions in Lyme disease at various resolution scale. Correlative light and electron microscopy, atomic force microscopy-based single-molecule force spectroscopy and solution nuclear magnetic resonance have been used to shed light on the underlying mechanisms associated with Lyme disease Borrelia infection. Specifically, the key molecular players and interactions responsible for the variance in the pathogenicity and disease outcome of Borrelia species have been studied. The rationale behind such studies was highlighted by review articles, which are part of the thesis.
New diagnostic and therapeutic aspects of inflammatiory cardiomyopathy

Introduction: Inflammatory cardiomyopathy (DCMi) represents a non-familial form of dilated cardiomyopathy (DCM) and endomyocardial biopsy (EMB) is crucial for its diagnosis. Aims: To assess the prevalence of DCMi in patients with DCM of unclear origin, to evaluate the significance of serological tests for antibodies against infectious cardiotrophic agents and to analyze the effect of specific therapy guided by EMB results. Methods: EMB was performed in 56 subjects (mean age 52 ± 10 years) with DCM of unclear etiology and left ventricular (LV) ejection fraction (EF) < 40% with a history of heart failure less than 1 year. EMB samples were analyzed by immunohistochemistry, polymerase chain reaction (PCR) and electron microscopy. Results: Immunohistochemical examination revealed myocardial inflammation in 26 patients (46%), the PCR method detected genome of microbial agents in 32 patients (57%). Electron microscopy showed the presence of particles of microbial agents in 41 patients (73%). Serological blood tests found no IgM antibody positivity against any of the investigated microbial agents. Targeted antibiotic therapy in patients with evidence of Borrelia burgdorferi (Bb) genome in the EMB led to a reduction in LV size, improvement of LV EF and alleviate symptoms of heart failure. Gene expression and infectivity of \kur{Borrelia afzelii} in the course of tick feeding

Borrelia afzelii differential gene expression in the course of tick blood-feeding, and during chronic infection in mice was studied. Temperature effect on B. afzelii gene expression and infectivity was investigated. Infection rates of mice immunized with B. afzelii tick gut antigen at various stages of tick blood-intake were analyzed.
